Given the failure of this expedition, Pius V brought the Spanish and Venetian plenipotentiaries to try to take effective measures against the Turkish expansion in the Mediterranean. The negotiations were delayed nearly a year due to the disparate interests and projects of Venetian and Spanish. Finally on May 25 1571, Spain, the Papal States, Venice and Malta signed the capitulations to form the Holy League. The agreement that was reached, promised to Philip II to contribute half of the soldiers and money, both Pius V with 1 / 6 and Venice with 2 / 6.

The fleet comprised 134 ships Venetian (6 galeazas, 106 galeras, 2 ships and 20 frigates), 164 Spanish boat (90 galleys, 24 ships and 50 frigates and Berganti os), 18 ships of the papacy (12 galleys and frigates 6) and 9 galleys of Malta.
The Spanish soldiers totaled 20,000, the Pope's 2000, 8000 and the Venetians few knights of the Order of Malta. The fleet was led by Juan de Austria. Marco Antonio Colonna, Condestable vassal of Naples and of Spain, was the admiral of the pope and Sebastian Veniero the Venetian ships.
On August 29, Bishop Odescalco reached Messina, gave the apostolic blessing on behalf of the Pope and granted indulgences of special Jubilee Cross and the whole army. On September 15th, John of Austria ordered the departure of the fleet anchorage at Corfu on 26, while a flotilla headed by Gil de Andrade explored the area.
